目的观察结直肠癌组织中肿瘤干细胞标志物CD133表达变化,并探讨其意义。方法采用免疫组化法检测60例手术切除的结直肠癌组织中的CD133、survivin、Ki67,并分析三者之间的相关性及其与患者性别、年龄、肿瘤的部位和大小、病理分级、侵袭深度、淋巴结转移、pTNM分期之间的关系。结果本组CD133阳性表达率为50%(30/60);CD133的表达与淋巴结转移和pTNM分期有关(P均<0.01),与病理分级有关(P<0.05),与其他临床病理参数无关(P均>0.05);survivin仅与肿瘤病理分级有关(P<0.05),Ki67与各项临床病理参数无关(P均>0.05)。本组CD133与survivin、Ki67的表达呈正相关(r分别为0.269、0.466,P均<0.05),survivin与Ki67的表达呈正相关(r为0.378,P<0.01)。结论结直肠癌组织中存在CD133阳性肿瘤细胞,即肿瘤干细胞。肿瘤干细胞在结直肠癌的发生、发展中起重要作用,检测肿瘤干细胞标志物CD133有助于判断肿瘤的恶性程度和预后。
Objective To observe the expression of tumor stem cell marker CD133 in colorectal cancer and to explore its significance. Methods The expressions of CD133, survivin and Ki67 in 60 resected colorectal cancer tissues were detected by immunohistochemistry. The correlations among the three were analyzed. The correlations were also analyzed with the gender, age, tumor location and size, pathological grade, Invasion depth, lymph node metastasis, pTNM staging. Results The positive expression rate of CD133 in this group was 50% (30/60). The expression of CD133 was correlated with lymph node metastasis and pTNM stage (all P <0.01), but not with other clinical pathological parameters (P <0.05) P> 0.05). Survivin was only associated with tumor pathological grade (P <0.05). Ki67 had no correlation with clinicopathological parameters (P> 0.05). The expression of survivin and Ki67 in this group was positively correlated (r = 0.269,0.466, P <0.05, respectively). There was a positive correlation between the expression of survivin and Ki67 (r = 0.378, P <0.01). Conclusion There are CD133 positive tumor cells in colorectal cancer tissues, namely tumor stem cells. Cancer stem cells play an important role in the occurrence and development of colorectal cancer. Detection of tumor stem cell marker CD133 can help to determine the malignancy and prognosis of cancer.
