目的 :观察大黄素对人肾成纤维细胞增殖及IL - 6分泌的影响。方法 :在人肾成纤维细胞体外培养的基础上 ,采用3 H -TdR掺入法和ELISA方法分别检测了不同浓度大黄素诱导下人肾成纤维细胞增殖及IL - 6的分泌。结果 :体外人肾成纤维细胞具有自发增殖过程 (P <0 .0 1) ,TMP能促进其增殖及IL - 6的分泌 (P <0 .0 1) ,大黄素则以剂量依赖方式降低了人肾成纤维细胞3 H -TdR掺入率和IL - 6的产生 (r =0 .99,P <0 .0 1;r =0 .99,P <0 .0 1)。结论 :大黄素能抑制人肾成纤维细胞增殖和IL - 6的产生 ,这一结果为大黄素的临床应用提供了部分实验依据
Objective: To observe the effects of emodin on proliferation of human renal fibroblasts and secretion of IL-6. METHODS: Based on the in vitro culture of human renal fibroblasts, 3 H-TdR incorporation and ELISA were used to detect the proliferation of human renal fibroblasts and the secretion of IL-6 induced by different concentrations of emodin. RESULTS: In vitro human renal fibroblasts had a spontaneous proliferation (P < 0.01), TMP promoted its proliferation and IL-6 secretion (P <0.01), and emodin decreased in a dose-dependent manner. 3 H-TdR incorporation rate and IL-6 production in human renal fibroblasts (r = 0.99, P < 0.01; r = 0.99, P < 0.01). Conclusion: Emodin can inhibit the proliferation of human renal fibroblasts and the production of IL-6. This result provides some experimental basis for the clinical application of emodin.
