目的从中药复方“前愈”中提取分离有效部位群并将其进行纯化,建立各有效部位(总生物碱,总黄酮,总多糖,总皂苷)的含量测定方法。方法采用溶剂法分离纯化总生物碱有效部位;溶剂法结合聚酰胺树脂柱层析法分离纯化总黄酮有效部位;水溶醇沉法分离纯化总多糖有效部位;采用溶剂法结合大孔树脂柱层析法分离纯化总皂苷有效部位。应用紫外分光光度法分别测定分离提取液及纯化液中各有效部位的含量。结果提取分离所得总生物碱,总黄酮,总皂苷,总多糖含量分别为2.07%,8.87%,9.46%,2.82%。纯化后各部位含量分别为50.52%,57.04%,74%,67.8%。结论本实验确定的分离纯化方法操作简便合理,可显著提高“前愈”复方中各有效部位的含量。
OBJECTIVE To extract and purify the active fractions from the compound Chinese medicine “Qian Yu”, and to establish a method for the determination of the effective fractions (total alkaloids, total flavonoids, total polysaccharides and total saponins). Methods The effective fractions of total alkaloids were isolated and purified by solvent method. The active fractions of total flavonoids were isolated by solvent method and polyamide resin column chromatography. The effective fractions of total polysaccharides were separated and purified by water-alcohol precipitation method. Method to separate and purify the effective parts of total saponins. UV spectrophotometry were used to determine the content of each active site in the separated extract and purified solution respectively. Results The total alkaloids, total flavonoids, total saponins and total polysaccharides were 2.07%, 8.87%, 9.46% and 2.82%, respectively. After purification, the content of each part was 50.52%, 57.04%, 74%, 67.8% respectively. Conclusion The separation and purification methods identified in this experiment are simple and reasonable in operation, and can significantly increase the content of each active site in the “Qian Yu ” compound.
