骨髓细胞长期培养的形态学研究 粘着性基质细胞的特性及其在维持血源性干细胞增生方面的互相作用

在骨髓的长期培养中,粘着性基质细胞对血源性千细胞的增生起着支持作用。这些基质细胞及它们的互相作用是通过扫描电镜及透射电镜与功能研究中发现的。在已形成的粘着性基质层上种植培养物,就象在土壤上播种一样,三周后加上另一些骨髓细胞悬液。细胞群落的出现证明血源性千细胞的持续增生。基质主要由两种细胞组成,即巨噬细胞和上皮样细胞。巨噬细胞较小,直径10—15μm,能吞噬乳胶及碳粒,含有溶酶体;其表面不能用多价铁(PCF)染色。上皮样细胞较大,直径超过100μm,含有大量细长的粒线体,不能吞噬乳胶颗粒,细胞膜的表面能被PCF染色。在透射电镜下,上皮样细胞的外观,因细胞的发育状态和切面的方向是平行于基层还是垂直于基层而定。上皮样细胞有一种成熟的光谱,在光谱的两端是合成相及贮存相。在合成相内,细胞含有大量粗面内质网,而在贮存相内则含有大量的贮存颗粒。这两种相在垂直于基层的切片上最容易识别,合成细胞在表层,而成熟的贮存细胞则位于基层。巨噬细胞及上皮样细胞内均含有脂肪球,脂肪球数量及体积的增加与在培养液中加入的皮质醇有关。在本研究中未发现清楚的脂肪细胞。当上清液无细胞群落激活时,在培养的过程中发现了粒细胞的发生,这说明在粒细胞发生的诱导下细胞间发生了直接的互相作用或短周期的因子。巨噬细胞及上皮样细胞具有广泛的细胞间的互相作用,上皮样细胞通常完全包围巨噬细胞,两种细胞互相间均伸出细胞质突起。在鸡胚造血时内胚层与中胚层就出现这样的互相作用,这对培养中千细胞的生存是必需的。 In the long-term culture of bone marrow, adherent stromal cells play a supportive role in the proliferation of hematopoietic stem cells. These stromal cells and their interactions are found by scanning electron microscopy and transmission electron microscopy and functional studies. Cultures were planted on the formed adhesive matrix layer as if sown on the soil and three weeks later, additional myeloid cell suspensions were added. The appearance of cell colonies demonstrates the continued proliferation of hematopoietic stem cells. The stroma consists mainly of two types of cells, macrophages and epithelial cells. Macrophages smaller, 10-15 μm in diameter, can swallow latex and carbon particles, containing lysosomes; its surface can not be stained with polyvalent iron (PCF). Epithelioid cells larger, more than 100μm in diameter, contains a large number of elongated mitochondria, can not swallow latex particles, cell membrane surface can be PCF staining. Under TEM, the appearance of epithelioid cells depends on whether the cell’s developmental state and slice orientation are parallel to the grass roots or to the grass roots. Epithelioid cells have a mature spectrum, at both ends of the spectrum is the synthesis and storage phase. Within the synthesis phase, the cells contain a large amount of rough endoplasmic reticulum, while in the storage phase they contain large quantities of stored particles. The two phases are most easily identified on slices perpendicular to the basal lamina, with the synthetic cells on the surface and the mature storage cells on the basal layer. Both macrophages and epithelial cells contain fat globules. The increase in the number and volume of fat globules is related to the addition of cortisol in the culture fluid. No clear adipocytes were found in this study. When the supernatant was activated without cell community, the occurrence of granulocytes was found during culture, indicating that there was a direct interaction or a short cycle between the cells under the induction of granulosis. Macrophages and epithelioid cells have extensive intercellular interactions. Epithelial-like cells usually completely surround macrophages, and both cells project out of the cytoplasm. This interaction occurs between endoderm and mesoderm during hematopoiesis in chicken embryos, which is necessary for the survival of the stem cells in culture.
