目的:以黄连解毒汤为研究对象,探讨其采用不同相对截留分子质量聚醚砜(PES)超滤膜处理后HPLC特征图谱的变化。方法:用pes-5w,pes-3w,pes-2w,pes-1w,pes-6k的超滤膜处理黄连解毒汤离心液,应用特征图谱技术,分析经各级膜处理后黄连解毒汤成分的种类和含量情况;并测定其浊度、粘度等理化参数。结果:各级膜处理液的药根碱、小檗碱、黄芩素、汉黄芩素逐级变小明显,栀子苷有明显的逐级上升的现象;各药液的理化参数(pH、浊度、电导率)也呈规律性的变化;以离心液为参照,按膜相对分子质量由大到小排序样品的相似度依次为0.993,0.992,0.984,0.982,0.975。结论:本实验运用了膜技术这一较新的分离精制工艺,采用特征图谱全面地反映黄连解毒汤的成分变化,发现各药液成分在种类和含量方面有差异,并探讨了理化参数变化的原因。
OBJECTIVE: To investigate the changes of HPLC profiles of Huanglian Jiedu Decoction treated with polyethersulfone (PES) membranes with different relative molecular mass cut offs. Methods: The extract of Huanglian Jiedu decoction was treated with ultrafiltration membrane of pes-5w, pes-3w, pes-2w, pes-1w and pes-6k. Species and content of the situation; and determine the turbidity, viscosity and other physical and chemical parameters. Results: The jatrorrhizine, berberine, baicalein and wogonin of the membrane-treating solution at all levels gradually became smaller and smaller, and the jasminoidin obviously increased gradually. The physicochemical parameters (pH, turbidity Degrees, conductivity) also showed a regular change; Centrifugal fluid as a reference, according to the membrane relative molecular mass descending order of similarity of samples were 0.993,0.992,0.984,0.982,0.975. Conclusion: In this experiment, membrane technology, a relatively new separation and purification process, was used to reflect the composition changes of Huanglian Jiedu Decoction comprehensively by using the characteristic map. It was found that there were differences in the types and contents of the constituents of the solution and the changes of physicochemical parameters the reason.
