目的:评价 MTT法测定肿瘤细胞体外药物敏感性的临床应用。方法:对75例胃癌、55例肠癌、55例膀胱癌及6例舌癌进行肿瘤细胞分离、培养,以MTT法对其体外药物敏感性进行检测。结果:对胃癌、肠癌、膀胱癌敏感度较高的药物与临床经验用药基本相符。75例胃癌病人体外药敏结果和体内疗效总符合率为77.3%,其中MTT敏感组病人总有效率与经验化疗组相比有显著性差异(P<0.01)。MTT法对新药的临床应用也有较好的指导作用。结论:MTT法对于临床合理用药,实现肿瘤病人的个体化化疗具有重要的价值。
OBJECTIVE: To evaluate the clinical application of MTT assay for measuring drug sensitivity of tumor cells in vitro. Methods: Tumor cells were isolated and cultured in 75 cases of gastric cancer, 55 cases of colorectal cancer, 55 cases of bladder cancer and 6 cases of tongue cancer. MTT assay was used to detect the drug sensitivity in vitro. Results: Drugs with high sensitivity to gastric cancer, colon cancer and bladder cancer were basically consistent with clinical experience. The total coincidence rate of in vitro drug sensitivity and in vivo efficacy in 75 patients with gastric cancer was 77.3%. The total effective rate of patients in the MTT sensitive group was significantly different from that in the chemotherapy group (P<0.01). The MTT method also has a good guiding role in the clinical application of new drugs. Conclusion: The MTT method is of great value for the rational use of drugs in the clinic and for individualized chemotherapy for cancer patients.
