目的:研究喉鳞状细胞癌组织中信号转导和转录激活因子3(Stat3)及其靶基因细胞周期蛋白D1(Cyclin D1)的表达情况并探讨其临床意义。方法:应用Western blot和RT-PCR方法,研究喉鳞状细胞癌组织中Stat3、Cyclin D1蛋白和mRNA的表达情况以及与喉鳞状细胞癌临床病理因素的关系。结果:喉鳞状细胞癌组织中Stat3、Cyclin D1蛋白和mRNA的表达水平明显高于正常喉黏膜组织(P<0.01)。Stat3、Cyclin D1蛋白表达水平与喉鳞状细胞癌临床分期、分化程度及淋巴结转移有关(P<0.01);在喉鳞状细胞癌组织中Stat3蛋白、mRMA与CyclinD蛋白、mRNA的表达均呈正相关(r=0.564、0.552,均P<0.01)。结论:Stat3信号转导通路在喉鳞状细胞癌的发生、发展过程中起重要作用,检测喉鳞状细胞癌组织中Stat3及其靶基因Cyclin D1的表达可以反映肿瘤的恶性程度。
Objective: To investigate the expression of signal transducer and activator of transcription 3 (Stat3) and its target gene cyclin D1 in laryngeal squamous cell carcinoma and to investigate its clinical significance. Methods: The expressions of Stat3, Cyclin D1 protein and mRNA in laryngeal squamous cell carcinoma and their clinicopathological factors were studied by Western blot and RT-PCR. Results: The expression of Stat3, Cyclin D1 protein and mRNA in laryngeal squamous cell carcinoma was significantly higher than that in normal laryngeal mucosa (P <0.01). The expression of Stat3 and Cyclin D1 was correlated with the clinical stage, differentiation and lymph node metastasis of laryngeal squamous cell carcinoma (P <0.01). Stat3 protein and mRMA were positively correlated with the expression of CyclinD protein and mRNA in laryngeal squamous cell carcinoma (r = 0.564, 0.552, all P <0.01). Conclusion: Stat3 signal transduction pathway plays an important role in the occurrence and development of laryngeal squamous cell carcinoma. Detecting the expression of Stat3 and its target gene Cyclin D1 in laryngeal squamous cell carcinoma can reflect the malignant degree of tumor.
