基于抗骨质疏松作用的淫羊藿拆分组分的筛选研究

目的:研究淫羊藿及其拆分组分对维甲酸致骨质疏松模型大鼠的影响,筛选最有效组分。方法:考察淫羊藿各组分对维甲酸致骨质疏松模型大鼠血清中Ca~(2+)、P~(3+)、ALP、Str ACP、BCG、E2的含量变化以及对骨钙、骨磷、双侧腿骨骨质量/体质量比、单侧股骨灰质量/干质量比的影响,并对股骨进行骨组织病理学观察。结果:淫羊藿30%乙醇组分可显著增加模型大鼠骨钙、骨磷含量和血清E2水平,降低血清ALP、Str ACP、BCG及磷含量,显著提高双侧腿骨质量/体质量比和单侧股骨灰质量/干质量比;淫羊藿全成分可显著增加骨钙、双侧股骨骨质量/体质量比和单侧股骨灰质量/干质量比,降低血清ALP及磷含量;骨组织形态学观察结果显示,除阳性对组外,淫羊藿30%乙醇段对骨病理形态改善最明显,可明显增加骨小梁的密度和间隙,仅局部骨壁呈间断性松质骨结构;综合各指标得出各组分抗骨质疏松作用大小依次为:淫羊藿30%乙醇段>淫羊藿全成分>淫羊藿水段>淫羊藿95%乙醇段>淫羊藿多糖。结论:淫羊藿各拆分组分中淫羊藿30%乙醇段在抗骨质疏松方面作用最显著,淫羊藿全成分也有较好的抗骨质疏松作用,但效果不如淫羊藿30%乙醇段,淫羊藿水段和淫羊藿95%乙醇段对骨质疏松可能有一定的预防作用,淫羊藿多糖的抗骨质疏松作用不明显。 Objective: To study the effects of epimedium and its fractions on rat model of osteoporosis caused by retinoic acid and to screen the most effective components. Results: Epimedium 30% ethanol could significantly increase the content of bone calcium and osteoporosis, serum E2 level and ALP, Str ACP, BCG and phosphorus in model rats, and significantly increase the ratio of bilateral leg mass to body mass And unilateral femoral gray mass / dry mass ratio; Epimedium the whole component can significantly increase bone calcium, bilateral femur bone mass / body mass ratio and unilateral femoral gray mass / dry mass ratio, lower serum ALP and phosphorus content; bone Histomorphological observation showed that except for the positive group, Epimedium 30% ethanol group had the most obvious improvement on the pathological morphology of bone, which could obviously increase the density and the gap of trabecular bone. Only the part of the bone wall showed discontinuous cancellous bone structure The results showed that the anti-osteoporosis effect of each component was as follows: Epimedium 30% ethanol> Epimedium all components> Epimedium water> Epimedium 95% ethanol> Epimedium polysaccharides . CONCLUSION: Epimedium 30% ethanol fraction is the most effective anti-osteoporosis component in Epimedium. Epimedium also has the best anti-osteoporosis effect, but the effect is not as good as Epimedium 30 % Ethanol, Epimedium water and Epimedium 95% ethanol may have a preventive effect on osteoporosis, Epimedium polysaccharide has no significant anti-osteoporosis.
