本文报道了大鼠腺垂体细胞单层培养的方法,经胰蛋白酶、DNA酶、胰蛋白酶抑制剂以及EDTA和神经氨酸酶先后处理的腺垂体细胞在24孔塑料培养板上生长良好,功能反应灵敏、稳定。LH分泌与细胞密度(0.25~2.0×10~6个/孔)呈高度相关。培养的细胞对GnRH(10~(-10)~10~(-6)mol/L)及下丘脑正中隆起提取物的反应良好,LH分泌量与刺激物浓度高度相关,最低有效刺激量分别为10~(-10)mol/L和0.00625“当量”。本法可作为下丘脑释放激素生理活性的检测系统,并可用于腺垂体机能调节的生理学,病理生理学和药理学研
This article reports the method of rat pituitary cell monolayer culture. The adenohypophyseal cells treated by trypsin, DNase, trypsin inhibitor, EDTA and neuraminidase grew well on 24-well plastic culture plate. Functional response Sensitive and stable.
