目的从低转移性人成骨肉瘤细胞系SOSP-9607中筛选出高转移亚群,并且建立人成骨肉瘤裸鼠转移模型。方法采用裸鼠腹腔传代的方法,获得腹水型自发性肺转移细胞,经体外培养和体内再次接种后获得高转移亚群。应用细胞计数法、染色体显示法、流式细胞术及裸鼠体内实验法研究细胞的生物学特性。结果筛选出的高转移亚群SOSP-M在转移分析实验中达到100%肺转移率,除肺转移外还出现其他器官的转移。其染色体为50余条,保持人类染色体核型。细胞周期中S期细胞所占比例为48.5%。SOSP-M的细胞形态、染色体数目、体外及体内生长速度均与母系SOSP-9607有很大差别。结论SOSP-M是一个腹水型高转移细胞亚群。
Objective To screen high metastatic populations from the low metastatic human osteogenic sarcoma cell line SOSP-9607 and establish a human metastatic model of osteosarcoma in nude mice. Methods Ascites spontaneous pulmonary metastatic cells were obtained by intraperitoneal passage of nude mice. High metastatic populations were obtained after in vitro culture and reinoculation in vivo. The cell biological characteristics were studied by cell counting, chromosome display, flow cytometry, and nude mice in vivo experiments. Results The high metastatic subsets of selected SOSP-M achieved a 100% lung metastasis rate in metastasis assays, with the exception of lung metastases and other organ metastases. Its chromosomes are more than 50, keeping human karyotypes. The proportion of cells in the S phase in the cell cycle is 48.5%. The cell morphology, chromosome number, in vitro and in vivo growth rate of SOSP-M were significantly different from that of maternal SOSP-9607. Conclusions SOSP-M is a subpopulation of ascites-type highly metastatic cells.
