目的检测颅内动脉瘤中细胞外基质相关基因的表达谱,探讨其在颅内动脉瘤发病机制中的作用。方法利用Affymetrix公司的人类基因组基因芯片U133A分别检测3例颅内动脉瘤组织和3例正常颅内动脉组织中的基因表达谱,用生物信息学软件进行比较分析,对差异表达基因进行功能分类,对其中的细胞外基质相关基因进行分析。结果与正常颅内动脉组织相比,颅内动脉瘤中有383个基因的差异表达水平达10倍以上;在这383个基因中,有23个基因与细胞外基质有关,其中11个基因的表达水平上调,12个基因的表达水平下调。结论细胞外基质相关基因的异常表达可能参与了颅内动脉瘤的形成和发展。
Objective To detect the expression profiles of extracellular matrix related genes in intracranial aneurysms and to explore its role in the pathogenesis of intracranial aneurysms. Methods Affymetrix human genomic microarray U133A was used to detect the gene expression profile in 3 cases of intracranial aneurysm and 3 cases of normal intracranial artery. Bioinformatics software was used to analyze the gene expression profiles of differentially expressed genes. One of the extracellular matrix-related genes were analyzed. Results Compared with normal intracranial arteries, 383 differentially expressed intracranial aneurysms were over 10-fold. Of the 383 genes, 23 were associated with extracellular matrix, of which 11 The expression level is up-regulated, the expression level of 12 genes is down-regulated. Conclusion The abnormal expression of extracellular matrix related genes may be involved in the formation and development of intracranial aneurysms.
