PPARγ2的Pro12Ala变异与肥胖、糖尿病的关联研究

目的 探讨过氧化物酶体增殖体激活受体γ2 (PPARγ2 )的Pro12Ala变异及其与肥胖或糖尿病之间的关系。方法 研究对象按BMI分为正常体重组 (91例 )与超重肥胖组 (14 0例 ) ;按WHO口服糖耐量检测标准分为糖耐量正常组 (10 3例 )、糖耐量减低组 (5 0例 )、糖尿病组 (78例 )。运用PCR以及限制性酶切鉴定方法检测PPARγ2Pro12Ala基因多态性 ,并测定血浆抗素 (Resistin)水平等指标。结果 PPARγ 2基因型Ala等位基因频率 (qA)在正常体重组为 3 85 % ,超重肥胖组为 3 93% ,基因型频率和等位基因频率在这两组之间没有显著性差异 (P >0 0 5 )。在PPARγ 2的不同基因型之间血浆抗素水平有显著性差异 (P <0 0 5 )。PPARγ 2基因Ala等位基因频率 (qA)在糖耐量正常组、糖耐量减低组与糖尿病组分别为 3 88%、4 0 0 %、3 85 % ,在这三组之间没有显著性差异 (P >0 0 5 ) ,在糖耐量正常组、糖耐量减低组、糖尿病组中PA +AA基因型者比PP基因型者血浆抗素水平要低 ,只是在糖耐量减低组PPARγ2基因Pro12Ala的不同基因型之间血浆抗素水平差异没有显著性 (P >0 0 5 )。结论 PPARγ2的Pro12Ala变异与肥胖、糖尿病无关 ,但PPARγ2的Pro12Ala变异可能与抗素表达、机体胰岛素敏感性有关。 Objective To investigate the Pro12Ala variation of peroxisome proliferator activated receptor γ2 (PPARγ2) and its relationship with obesity or diabetes mellitus. Methods The subjects were divided into normal weight group (91 cases) and overweight and obesity group (140 cases) by BMI. According to WHO oral glucose tolerance test, the subjects were divided into normal glucose tolerance group (103 cases), impaired glucose tolerance group Cases), diabetic group (78 cases). The polymorphisms of PPARγ2Pro12Ala gene were detected by PCR and restriction enzyme digestion, and the levels of plasma resistin (Resistin) were measured. Results The frequency of allele Ala of PPARγ 2 genotype was 85% in normal weight group and 3 93% in overweight obesity group. There was no significant difference in genotype frequency and allele frequency between the two groups (P > 0 0 5). There was a significant difference in the levels of plasma plasmids between different genotypes of PPARγ 2 (P <0 05). The Ala allele frequency (qA) of PPARγ 2 gene was 3 88%, 400%, 3 85% respectively in the normal glucose tolerance group, the impaired glucose tolerance group and the diabetic group. There was no significant difference between these three groups P <0.05). In the normal glucose tolerance group, the glucose tolerance group and the diabetic group, PA + AA genotype had lower level of plasma anti-angiotensin system than that of PP genotype, except for the difference of PPARγ2 gene Pro12Ala in impaired glucose tolerance group There was no significant difference in plasma levels of antigens between genotypes (P> 0.05). Conclusion The mutation of Pro12Ala in PPARγ2 is not related to obesity and diabetes. However, the mutation of Pro12Ala in PPARγ2 may be related to the expression of antibiotics and the insulin sensitivity.
