目的 观察血管紧张素 ( Ang )在体外对自发性高血压以及正常血压两种乳鼠左心室成纤维细胞( CFB)的促生长作用并通过应用 Ang 受体 型 ( AT1 R)抑制剂——洛沙坦以探讨 Ang 的作用途径。方法 酶解法制备乳鼠左心室 CFB,分为 ( 1)对照组 (不加任何药物 ) ;( 2 )不同浓度 Ang ( 10 - 1 0 ~ 10 - 6 )组 :( 3) 10 - 6 M洛沙坦组 ;( 4) 10 - 8MAng +不同浓度洛沙坦 ( 10 - 8~ 10 - 6 M)组。应用 3H- Td R、 3H- L eu掺入法测定细胞 DNA和蛋白质合成率。结果 Ang 使 CFB3H- Td R、 3H- L eu掺入率增加 ,呈剂量依赖性。洛沙坦可抑制 Ang 的作用。结论 Ang 在体外具有经细胞表面的 AT1 R促进 CFB增殖的作用
Objective To investigate the effects of angiotensin (Ang) on the growth of left ventricular fibroblasts (CFB) in spontaneously hypertensive rats and normotensive hypertensive rats in vitro and the effects of Angiotensin Ⅱ Sartan to explore the role of Ang. Methods The left ventricular CFB of neonatal rats was prepared by enzymolysis and divided into (1) control group (without any drug); (2) Angiotensin II (10 - 10 ~ (4) 10 - 8 MAG + different concentrations of losartan (10 - 8 ~ 10 - 6 M) group. Cell DNA and protein synthesis rates were determined by 3H-TdR, 3H-L eu incorporation. Results Ang increased the incorporation rate of CFB3H-TdR, 3H-L eu in a dose-dependent manner. Losartan can inhibit the role of Ang. Conclusion Ang in vitro with the cell surface of AT1 R promote CFB proliferation
