目的观察mPEG-SPA(甲氧基聚乙二醇-琥珀酰亚胺丙酸酸酯)对人红细胞RhD血型抗原修饰的效果,并探讨mPEG-SPA修饰的可行性。方法采用mPEG-SPA修饰人红细胞血型RhD抗原,并观察其对红细胞形态、结构和功能的影响。结果 2.0 mmol/L的mPEG-SPA在pH8.0室温条件下可有效遮蔽红细胞表面RhD抗原,且与红细胞结合牢固,对红细胞的形态、渗透脆性、自身溶血率、红细胞膜胆固醇含量、乙酰胆碱酯酶活性、Na+-K+ATP酶活性、2,3-DPG含量、血沉值、变形指数和常规指标均无明显影响。结论采用2.0 mmol/L mPEG-SPA遮蔽人红细胞RhD抗原获得了初步成功。
Objective To investigate the effect of mPEG-SPA (methoxypolyethylene glycol-succinimidyl propionate) on the modification of RhD blood group antigens in human erythrocytes and to explore the feasibility of mPEG-SPA modification. Methods mPEG-SPA was used to modify the blood group RhD antigen of human erythrocytes, and its effect on the morphology, structure and function of erythrocytes was observed. Results The 2.0 mmol / L mPEG-SPA could effectively block the RhD antigen on the surface of erythrocytes at room temperature of 8.0, and could bind with erythrocytes firmly. The morphology, osmotic fragility, hemolysis, erythrocyte membrane cholesterol, acetylcholinesterase Activity, Na + -K + ATPase activity, 2,3-DPG content, erythrocyte sedimentation rate, deformation index and conventional indicators had no significant effect. Conclusion 2.0 mmol / L mPEG-SPA was used to mask the RhD antigen of human erythrocytes.
