应用逆转录聚合酶链反应(RT-PCR)技术检测肝星状细胞(HSC)中5个生长抑素受体(SSTR)亚型的表达情况,从侧面探索生长抑素影响肝纤维化的作用机制。针对试验结果中SSTR的表达情况,采用异硫氰酸荧光素(FITC)标记的奥曲肽(OCT)作用于体外培养的HSC,然后在共聚焦显微镜下观察活细胞中奥曲肽的分布情况,从形态学上证实SSTR的存在。
The expression of 5 somatostatin receptor (SSTR) subtypes in hepatic stellate cells (HSC) was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and the effect of somatostatin on hepatic fibrosis mechanism. According to the results of SSTR expression, octreotide fluorescein isothiocyanate (FITC) -labeled octreotide (OCT) was used to in vitro cultured HSC. Then the distribution of octreotide in living cells was observed under confocal microscope. Confirmed the existence of SSTR.
