目的:考察不同提取方法中法半夏对黄连中盐酸小檗碱溶出率的影响。方法:选择复方葶苈泻肺利水合剂为考察对象,分别以水和乙醇为溶媒,采用方中法半夏、黄连、益母草、半边莲合提取及缺法半夏其余3味药材提取2种方法提取,比较提取液中盐酸小檗碱含量。以盐酸小檗碱提取量为指标,通过正交试验考察乙醇体积分数、提取时间、提取次数及料液比对复方葶苈泻肺利水合剂中4味药材提取工艺的影响。采用HPLC测定盐酸小檗碱含量,流动相乙腈-0.05 mol·L-1磷酸二氢钾溶液(50∶50)(每100 m L中加十二烷基硫酸钠0.4 g,以磷酸调节p H 4.0),检测波长345 nm。结果:以水为溶媒,2种方式提取的盐酸小檗碱含量差别较大,合提与缺法半夏提取时盐酸小檗碱平均质量分数分别为9.20,13.53 mg·g-1;以乙醇为溶媒,2种方式提取的盐酸小檗碱含量差别较小,平均质量分数分别为35.15,36.12 mg·g-1。4味药材的最佳提取工艺为加10倍量70%乙醇回流提取2次,每次1.5 h;盐酸小檗碱提取量37.92 mg·g-1。结论:以水为溶媒,法半夏与黄连合提将导致盐酸小檗碱含量显著降低;以乙醇为溶媒,法半夏与黄连合提对盐酸小檗碱溶出率影响较小,且在乙醇溶液中盐酸小檗碱提取率远高于水提法。优选的提取工艺稳定可行、重复性好,可用于复方葶苈泻肺利水合剂的工业化生产。
OBJECTIVE: To investigate the effects of different extraction methods of Chinese Pinellia Rhizoma on the dissolution rate of Berberine Hydrochloride in Coptis. Methods: Compound 葶 苈 Xie Fei Li Shui Mixture was selected as the object of study, respectively, water and ethanol as the vehicle, the use of Fangzhong France Pinellia, Coptis, Motherwort, Elaeagnus Amomum extract and lack of Banxiaxia the remaining three herbs extracted two ways Extract, compare the content of berberine hydrochloride in the extract. Taking the extraction amount of berberine hydrochloride as an index, the effects of ethanol volume fraction, extraction time, extraction times and the ratio of solid to liquid on the extraction technology of 4 herbs were studied by orthogonal test. The content of berberine hydrochloride was determined by HPLC. The mobile phase consisted of acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate solution (50:50) (0.4 g sodium dodecyl sulfate per 100 mL, phosphate 4.0), detection wavelength 345 nm. Results: The content of berberine hydrochloride extracted by two ways was different with water as solvent. The average content of berberine hydrochloride was 9.20 and 13.53 mg · g-1, As the vehicle, the content of berberine hydrochloride extracted by the two methods is quite different with the average mass fractions of 35.15 and 36.2 mg · g-1.4, respectively. The optimum extraction process is the addition of 10 times the amount of 70% ethanol reflux extraction 2 Times, each time 1.5 h; extraction of berberine hydrochloride 37.92 mg · g-1. Conclusion: With water as solvent, the combination of Pinellia and Coptis will lead to the significant decrease of the content of Berberine Hydrochloride. With ethanol as solvent, the effect of Pinellia and Coptis Extract has little effect on the dissolution rate of Berberine Hydrochloride, Berberine hydrochloride solution extraction rate is much higher than water extraction. The preferred extraction process is stable and feasible with good repeatability and can be used for industrial production of compound 葶 苈 Xie Li Li Shui Mixture.
