目的:探讨微小RNA-27a(mi RNA-27a)在宫颈上皮内瘤变(CIN)Ⅲ级患者和宫颈鳞癌患者血清及组织中的表达及意义。方法:选取2013年6月~2014年2月该院收治的宫颈鳞癌患者59例(宫颈癌组)及CINⅢ级患者37例(CINⅢ组)为研究对象,另选取同期宫颈正常者13例为对照组,分别提取3组患者血清及组织中的mi RNA,用实时荧光定量PCR法检测mi RNA-27a的表达情况。结果:宫颈癌组患者血清和组织中mi RNA-27a的表达量高于CINⅢ组及对照组(P<0.05),且其血清及组织中mi RNA-27a的表达呈正相关(r=0.782,P<0.05);不同分期及分级宫颈鳞癌患者的血清和组织中的mi RNA-27a表达水平比较,差异均无统计学意义(P>0.05)。结论:mi RNA-27a在宫颈鳞癌患者和CINⅢ级患者血清及组织中的表达水平均明显增高,提示其可能与宫颈鳞癌的发生及进展有关,可以考虑作为宫颈鳞癌的诊断指标。
Objective: To investigate the expression and significance of miRNA-27a in serum and tissues of cervical intraepithelial neoplasia (CIN) Ⅲ patients and cervical squamous cell carcinoma patients. Methods: From June 2013 to February 2014, 59 patients with cervical squamous cell carcinoma (CINC) and 37 patients with CINⅢ (CINⅢ) were enrolled in the study. Thirteen patients with normal cervix at the same period were selected as In the control group, the mi RNAs in the serum and tissues of the three groups were respectively extracted and the mi RNA-27a expression was detected by real-time fluorescence quantitative PCR. Results: The expression of miRNA-27a in serum and tissues of patients with cervical cancer was higher than that of CINⅢ and control (P <0.05), and the expression of miRNA-27a in serum and tissues was positively correlated (r = 0.782, P <0.05). There was no significant difference in the expression levels of miRNA-27a in serum and tissues between patients with different stages and grades of cervical squamous cell carcinoma (P> 0.05). Conclusion: The expression of mi RNA-27a in serum and tissues of patients with cervical squamous cell carcinoma and CINⅢlevels are significantly increased, suggesting that mi RNA-27a may be related to the occurrence and progression of cervical squamous cell carcinoma and may be considered as a diagnostic indicator of cervical squamous cell carcinoma.
