目的建立测定减肥通便类保健食品中芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚含量的高效液相色谱方法。方法保健食品样品以稀盐酸进行水解,通过液液萃取方法,采用岛津Shim-pack VP-ODS(4.6 mm×150 mm,5μm)色谱柱进行分离,以甲醇+0.1%磷酸溶液(85+15)为流动相,流速为1.0 ml/min,扫描波长范围200~400 nm,在波长254 nm处进行定性和定量分析。结果芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚分别在0.10~40、0.10~40、0.10~40、0.10~40、0.10~20μg/ml浓度范围内具有良好的线性关系;在低、中、高3个添加水平范围内的平均回收率为90.8%~103.5%,相对标准偏差(RSD)在0.20%~3.75%之间。结论该方法操作简便、结果准确,具有良好的重现性,技术参数指标符合保健食品理化分析的要求,可用于减肥通便类保健食品的质量控制。
OBJECTIVE To establish a HPLC method for determination of aloe-emodin, rhein, emodin, chrysophanol and physcion in health food for weight loss and catharsis. Methods The samples of health food were hydrolyzed with dilute hydrochloric acid and separated by liquid-liquid extraction using a Shimadzu Shim-pack VP-ODS column (4.6 mm × 150 mm, 5 μm). The samples were eluted with methanol + 0.1% phosphoric acid solution ) As the mobile phase at a flow rate of 1.0 ml / min and a scan wavelength range of 200-400 nm at a wavelength of 254 nm for qualitative and quantitative analysis. Results Aloe-emodin, rhein, emodin, chrysophanol and physcion had good linearity in the range of 0.10 ~ 40, 0.10 ~ 40, 0.10 ~ 40, 0.10 ~ 40 and 0.10 ~ 20μg / The average recoveries were 90.8% ~ 103.5% with RSDs of 0.20% ~ 3.75% at the three levels of low, medium and high levels. Conclusion The method is simple, accurate and reproducible. The technical parameters meet the requirements of physical and chemical analysis of health food and can be used for the quality control of health food.
