Preclinical studies Adriamycin is an anthracycline antibiotic with hydroxyl on the 14th carbon of daunorubicin. Adriamycin biological activity is related to its ability to specifically bind to DNA between adjacent base pairs inserted into a double helix. It disrupts the properties of stereochemical template sequences, and enzymes that inhibit DNA replication and transcription. The results of cell-free system studies are consistent with those obtained in vivo and in cell culture (ie, inhibition of precursor insertion into DNA and RNA, inhibition of mitosis, and chromosomal aberrations). Adriamycin is effective against mouse Ehrlich ascites carcinoma, lymphosarcoma C3H mice, rat Oberling-Guerin-Guerin myeloma, mouse sarcoma S-180 and leukemia L-1210.
