目的观察环孢霉素A(Cyclosporine A,CsA)和苦参碱(Matrine,MAT)联合对膀胱肿瘤T24/ADM细胞耐药性的逆转作用。方法以T24/ADM细胞为靶细胞,MTT法检测CsA对T24/ADM细胞的细胞毒效应及逆转作用。采用MTT法、流式细胞术分析CsA和MAT联合对T24/ADM细胞的逆转作用,观察CsA和MAT联合对T24/ADM细胞生长的抑制作用及检测T24/ADM细胞膜P-gp的表达情况。结果 CsA在非毒性剂量浓度4μg/ml以下能逆转细胞耐药。CsA和MAT单独应用均对T24/ADM细胞的耐药性有较强的抑制效应,两者联合呈协同效应。与T24/ADM耐药细胞组比较,T24/ADM+MAT组p-gp表达量降低(P<0.01),而T24/ADM+CsA组p-gp表达量无明显变化(P>0.05),但两者联合应用时作用大于两者单独作用之和。结论 CsA和MAT联合可协同逆转膀胱肿瘤T24/ADM细胞的耐药性,其作用机制可能为下调T24/ADM细胞膜P-gp的表达水平。
Objective To investigate the reversal effect of Cyclosporine A (CsA) and Matrine (MAT) on the drug resistance of bladder tumor T24 / ADM cells. Methods T24 / ADM cells were used as target cells. Cytotoxicity and reversal effect of CsA on T24 / ADM cells were detected by MTT assay. MTT assay was used to detect the reversal effect of CsA and MAT on T24 / ADM cells by flow cytometry. The inhibitory effect of CsA and MAT on the growth of T24 / ADM cells and the expression of P-gp in T24 / ADM cells were observed. Results CsA could reverse the cell resistance in the non-toxic dosage of 4μg / ml. Both CsA and MAT alone had a strong inhibitory effect on the drug resistance of T24 / ADM cells, showing a synergistic effect. The expression of p-gp in T24 / ADM + MAT group was significantly lower than that in T24 / ADM + ADM group (P <0.01), but not in T24 / ADM + CsA group (P> 0.05) The combined effect of the two is greater than the combined effects of both. Conclusion The combination of CsA and MAT can reverse the drug resistance of bladder cancer T24 / ADM cells. The mechanism may be that the expression of P-gp is down-regulated in T24 / ADM cell membrane.
