目的 探讨尼古丁对缺血后大鼠心脏去甲肾上腺素释放的影响及其机制。方法 以离体大鼠心脏缺血模型 ,测定尼古丁加入前后的去甲肾上腺素 (noradrenaline,NA)、神经肽Y(neuropeptideY ,NPY)及二羟苯基乙二醇 (dihydroxyphenylethyleneglycol,DOPEG)含量的变化 ,并探讨它们之间的关系。结果 (1 )非缺血条件下 ,低浓度尼古丁灌注液对去甲肾上腺素的释放无明显影响 ;(2 )在缺血 1 0min后 ,低浓度尼古丁灌注液组较对照组去甲肾上腺素及神经肽Y释放明显增加 ;(3)缺血 2 0min组与缺血 1 0min组相比 ,神经肽含量无明显变化 ,但去甲肾上腺素及DOPEG含量明显上升。结论 尼古丁促进缺血心脏以胞裂 (exocytosis)和非胞裂 (nonexcytosisin)方式释放去甲肾上腺素
Objective To investigate the effect and mechanism of nicotine on norepinephrine release from ischemic rat heart. Methods The changes of noradrenaline (NA), neuropeptide Y (NPY) and dihydroxyphenylethyleneglycol (DOPEG) in the rat cardiac ischemia model were determined before and after the addition of nicotine , And explore the relationship between them. Results (1) In the condition of non-ischemia, low concentration of nicotine perfusate had no significant effect on the release of norepinephrine; (2) After 10 minutes of ischemia, nicotine perfusion group of low concentration compared with noradrenaline control group Neuropeptide Y release increased significantly; (3) ischemic 20min group and ischemic 10min group, neuropeptide content no significant change, but norepinephrine and DOPEG content increased significantly. Conclusions Nicotine promotes the release of norepinephrine in the ischemic heart by both exocytosis and nonexcytosis
