目的 探讨增殖生长因子(PCNA、Ki-67)及癌基因蛋白(p53、ras p21、c-myc及c-erbB-2)在人肉瘤及移植癌中的表达特征、相关性及生物学特性.方法 用免疫组织化学ABC法检测增殖生长因子及癌基因蛋白产物在人肉瘤及移植瘤的42个组织样本中的表达.结果 人肉瘤及移植瘤组中Ki-67、PCNA表达率及p53蛋白过度表达率均高于对照组(P<0.05);而c-erbB-2呈现低表达.癌基因与抑癌基因有协同表达.Ki-67表达与PC-NA表达,核分裂相,%S及%S+G_2M密切相关;PCNA表达与核分裂相,%S密切相关;Ki-67和PCNA两者的表达均与DI不相关.结论 Ki-67和PCNA是判断肉瘤生长速率和进展的较理想的标记物;p53蛋白的过度表达在肉瘤细胞的增殖、发展中起重要作用;肉瘤的发生及进展与多种癌基因与抑癌基因的协同作用有关.
Objective To investigate the expression characteristics, correlation and biological characteristics of proliferating growth factor (PCNA, Ki-67) and oncogene proteins (p53, ras p21, c-myc and c-erbB-2) in human sarcoma and xenograft carcinoma. Methods Immunohistochemical ABC method was used to detect the expression of proliferative growth factor and oncoprotein in human tissue sarcoma and 42 tumor tissues. The results showed that Ki-67, PCNA expression and p53 protein were overexpressed in human sarcoma and transplanted tumors. The expression rate was higher than that of the control group (P<0.05); c-erbB-2 was lowly expressed. The oncogene and tumor suppressor gene had synergistic expression. Ki-67 expression and PC-NA expression, mitotic phase, %S and % S+G2M is closely related; PCNA expression is closely related to the mitotic phase, %S; neither Ki-67 nor PCNA expression is related to DI. Conclusion Ki-67 and PCNA are ideal markers for judging the growth rate and progression of sarcoma. The overexpression of p53 protein plays an important role in the proliferation and development of sarcoma cells. The occurrence and progression of sarcoma are related to the synergistic effects of various oncogenes and tumor suppressor genes.
