目的:研究当归补血汤对高糖条件下肾小球系膜细胞(GMC)增殖及其乙酰肝素酶(HPA)表达的影响,探讨其在糖尿病肾病(DN)防治中的意义。方法:①利用透射电镜观察各组中GMC的增殖情况;②采用免疫组织化学方法检测各组肾小球中HPA的表达。结果:高糖能明显促进肾小球GMC增殖,也相应的增加系膜细胞中HPA表达(P<0.05);而当归补血汤能够明显抑制GMC增殖和相应减少HPA在肾小球中的表达(P<0.05),且效果优于格列奇特组。结论:高糖可以加强GMC增殖及细胞中HPA表达,当归补血汤能够明显抑制GMC增殖及细胞中HPA表达,从而预防肾小球硬化的发生、发展和恶化,进而延缓DN的进展。
Objective: To study the effect of Dangguibuxuetang on the proliferation of glomerular mesangial cells (GMC) and its expression of heparanase (HPA) under high glucose conditions and its significance in the prevention and treatment of diabetic nephropathy (DN). Methods: (1) Transmission electron microscopy was used to observe the proliferation of GMC in each group; (2) Immunohistochemistry was used to detect the expression of HPA in glomeruli. Results: High glucose could promote the proliferation of glomerular GMC and increase the expression of HPA in mesangial cells (P <0.05), while Dangguibuxuetang could obviously inhibit the proliferation of GMC and correspondingly decrease the expression of HPA in glomeruli P <0.05), and the effect is better than gliclazide group. CONCLUSION: High glucose can enhance the proliferation of GMC and the expression of HPA in cells. Dangguibuxuetang can obviously inhibit the proliferation of GMC and the expression of HPA in cells, thereby preventing the occurrence, development and deterioration of glomerulosclerosis and thus slowing the progression of DN.
