[目的]研究海马内环境稳态在抑郁症中的作用。[方法]40只雌性Wistar大鼠分为对照组、模型组、蒸馏水组和中药组。ELISA法检测血清皮质醇、雌二醇,光镜和电镜观察海马CA3区形态学变化,免疫组化法检测海马脑源性神经生长因子和血管内皮生长因子表达。[结果]与对照组比较,模型组和蒸馏水组皮质醇水平显著升高,雌二醇水平明显下降,海马CA3区神经元损伤严重,脑源性神经生长因子和血管内皮生长因子表达明显降低,中药组上述指标变化不明显。[结论]抑郁状态下,海马内环境稳态被破坏,定志小丸具有一定的维持海马内环境稳态的作用。
[Objective] To study the role of homeostasis of hippocampus in depression. [Methods] Forty female Wistar rats were divided into control group, model group, distilled water group and TCM group. Serum cortisol, estradiol, light microscope and electron microscope were used to detect the morphological changes of hippocampal CA3 region by ELISA. The expression of brain-derived nerve growth factor and vascular endothelial growth factor were detected by immunohistochemical method. [Results] Compared with the control group, the cortisol level of model group and distilled water group were significantly increased, the estradiol level was significantly decreased, the neurons in hippocampal CA3 area were severely damaged, the expression of brain-derived neurotrophic factor and vascular endothelial growth factor were significantly decreased, Chinese medicine group changes in the above indicators is not obvious. [Conclusion] The homeostasis of hippocampus in the depression state is destroyed, and Dingzhi Xiaowan can maintain the homeostasis in the hippocampus.
