目的建立液相色谱-串联质谱(LC/MS/MS)法同时测定芒果苷和小檗碱在大鼠血浆中的浓度。方法以对乙酰氨基酚为内标,血浆样品采用沉淀蛋白法处理。用电喷雾离子化和正离子多离子反应监测(MRM)方式检测芒果苷和小檗碱。结果该方法芒果苷和小檗碱的线性范围分别为3.01~300.60 ng.mL-1和2.66~266.20 ng.mL-1;定量下限(LLOQ)分别为3.01 ng.mL-1和2.66 ng.mL-1;方法回收率分别在97.0%~103.2%和94.0%~101.1%;批内、批间变异系数均<12%。结论该方法准确、灵敏、特异、简便,适用于鼠血浆芒果苷和小檗碱浓度的同时测定。
Objective To establish a liquid chromatography-tandem mass spectrometry (LC / MS / MS) method for the simultaneous determination of mangiferin and berberine in rat plasma. Methods Acetaminophen was used as internal standard, and plasma samples were treated by precipitation protein method. Detection of mangiferin and berberine by electrospray ionization and positive ion multi-ion reaction monitoring (MRM). Results The linear range of this method was 3.01 ~ 300.60 ng.mL-1 and 2.66 ~ 266.20 ng.mL-1, and the limit of quantification (LLOQ) was 3.01 ng.mL-1 and 2.66 ng.mL -1. The recoveries were 97.0% ~ 103.2% and 94.0% ~ 101.1%, respectively. Conclusion The method is accurate, sensitive, specific and simple and suitable for the simultaneous determination of plasma mangiferin and berberine concentrations.
