淀粉样前体蛋白(amyloid precursor protein,APP)属于跨膜蛋白,最初于神经系统中发现且与阿尔茨海默病(Alzheimer’s disease,AD)的发病机制相关,随后外周组织中如上皮细胞、肝、心肌等也有发现.它能被多个分泌酶分解,全长APP及其产物均有许多功能,如分泌形式的APP对角质形成细胞具有促细胞增殖、运动和抗凋亡效应.但其突变、上调或下调可与许多疾病密切相关,如AD和黑素瘤.此综述主要介绍APP在皮肤生理功能和皮肤疾病的研究.“,”Amyloid precursor protein (APP),a transmembrane protein that was firstly found in the nervous system was associated with the pathogenesis of Alzheimer’s disease (AD).Then it was also expressed in peripheral tissues such as epithelial cells,liver and myocardium. It can be resolved by multiple secretases,and the full-length APP and its hydrolyzates have many functions. For example,secreted forms of APP have the effects of promoting cell proliferation,motility and anti-apoptosis on keratinocytes. However,its mutation,up-regulation or down-regulation was closely related to a variety of diseases such as AD and melanoma. This review focuses on physiological and pathological functions of APP in the skin.
