目的观察C57BL/6小鼠感染日本血吸虫后,体内细胞因子IL 22来源细胞的动态变化,并分析小鼠体内IL 22的主要来源。方法分别提取感染日本血吸虫0、3、5、8、13周的小鼠淋巴结和脾脏细胞,采用流式细胞术检测各个感染时期IL 22来源细胞的比例。结果感染小鼠的淋巴结和脾脏中,产生IL 22的细胞比例显著升高。在所有产生IL 22的细胞中,有80%左右为T细胞。产生IL 22的CD4-T细胞的比例在感染后3周内明显升高,而产生IL 22的CD4+T细胞的比例到感染第3周后才开始升高,至第8周显著增加。结论日本血吸虫感染小鼠的淋巴结和脾脏中,IL 22由T细胞和非T细胞两大群细胞产生,其中T细胞是细胞因子IL 22最主要的来源。产生IL 22的T细胞分为CD4-T细胞和CD4+T细胞两群,其中产生IL 22的CD4-T细胞在日本血吸虫感染早期迅速增加;而在感染进入慢性期后产生IL 22的CD4+T细胞大量增加。
Objective To observe the dynamic changes of IL-22-derived cells in C57BL / 6 mice infected with Schistosoma japonicum and to analyze the main sources of IL-22 in mice. Methods Lymph nodes and spleen cells of mice infected with Schistosoma japonicum were collected at 0, 3, 5, 8 and 13 weeks respectively. The proportion of IL-22-derived cells in each infection period was detected by flow cytometry. Results In the lymph nodes and spleens of infected mice, the proportion of cells producing IL-22 was significantly increased. About 80% of all IL-22 producing cells are T cells. The proportion of CD4-T cells producing IL-22 increased significantly within 3 weeks after infection, while the proportion of IL-22 producing CD4 + T cells began to increase after the third week of infection and increased significantly by the 8th week. Conclusion In the lymph nodes and spleens of mice infected with Schistosoma japonicum, IL 22 is produced by two large population of T cells and non-T cells, of which T cells are the most important source of cytokine IL 22. T cells that produce IL-22 are divided into two groups of CD4-T cells and CD4 + T cells, of which CD4-T cells that produce IL-22 rapidly increase in the early stage of Schistosoma japonicum infection; CD4 + T cells increased significantly.
