目的:研究雷奈酸锶对大鼠快速扩弓过程中腭中缝骨形态发生蛋白2(bone morphogenetic protein-2,BMP-2)表达的影响。方法:选取36只6周龄雄性Wistar大鼠,随机分为A、B、C 3组,每组12只,其中,A组为对照组。将机械扩弓装置置于B、C 2组大鼠上颌两侧第一、二磨牙之间,并加力100 g,A组不做处理。C组大鼠每天灌服600 mg/kg雷奈酸锶溶液,B组灌服相同剂量的生理盐水。分别于加力后4、7、10 d分批处死大鼠,用免疫组织化学染色法检测大鼠腭中缝组织中BMP-2的表达,并利用Image-pro plus粘结对切片吸光度进行分析,采用SPSS19.0软件包对数据进行统计学分析。结果:免疫组织化学染色显示,B组大鼠腭中缝组织中BMP-2的表达量在第4天时显著高于A组(P<0.05),而在第7天和第10天时与A组无显著差异(P>0.05);C组大鼠腭中缝组织中BMP-2的表达量在各个时间点均显著高于其余2组(P<0.05)。结论:雷奈酸锶能够促进大鼠快速扩弓过程中BMP-2的表达量和表达时长,可能促进了快速扩弓过程中腭中缝的新骨形成。
Objective: To investigate the effect of strontium ranelate on the expression of bone morphogenetic protein-2 (BMP-2) in rat palatal during rapid expansion. Methods: Thirty-six 6-week-old male Wistar rats were randomly divided into A, B and C groups, 12 in each group. A group was control group. The mechanical expansion device placed B, C 2 rats on both sides of the first molar between the two molar, and the force of 100 g, A group without treatment. Rats in group C were given 600 mg / kg of strontium ranelate solution daily, and group B was given the same dose of saline. The rats were sacrificed at 4, 7 and 10 d after burn-in respectively. The expression of BMP-2 in the rat palatal suture was detected by immunohistochemistry. The absorbance of the slices was analyzed by Image-pro plus. SPSS19.0 software package for statistical analysis of the data. Results: Immunohistochemical staining showed that the expression of BMP-2 in the palatal suture in group B was significantly higher than that in group A on the fourth day (P <0.05), but not on the seventh day and the tenth day (P> 0.05). The expression of BMP-2 in palatal suture in group C was significantly higher than that in other two groups at all time points (P <0.05). CONCLUSIONS: Strontium ranelate can promote the expression of BMP-2 and the expression of BMP-2 during the process of rapid expansion in rats, which may promote the new bone formation in the palatal midgut during rapid expansion.
