目的探讨枸杞多糖(LBP)对C6脑胶质瘤细胞株诱导的脑胶质瘤大鼠免疫功能的影响。方法将40只大鼠随机分为正常对照组、模型组、LBP组和顺铂组,每组10只。除正常对照组外,其余各组均制备大鼠脑胶质瘤动物模型。LBP组用LBP灌胃1次/d,顺铂组用顺铂灌胃1次/d,正常对照组和模型组用生理盐水灌胃1次/d,治疗时间共16周。ELISA法检测各组大鼠血清细胞因子中肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10)的水平;用流式细胞术检测各组大鼠外周血CD4~+CD25~+调节性T细胞(Tr)和CD8~+T细胞数量。结果与正常对照组相比,模型组大鼠外周血CD4~+CD25~+Tr数量增加而CD8~+T细胞的数量减少,血清IL-10水平增加而TNF-α水平降低(P<0.05);与模型组比较,LBP组和顺铂组大鼠外周血CD4~+CD25~+Tr数量减少而CD8~+T细胞数量增加,血清IL-10水平降低而TNF-α水平增加(P<0.05)。各组所测指标在8周与16周之间差异无统计学意义(P>0.05)。结论脑胶质瘤大鼠免疫功能降低,而LBP可增强实验性脑胶质瘤大鼠免疫功能,从而达到抗肿瘤的作用。
Objective To investigate the effect of LBP on immune function in glioma induced by C6 glioma cell lines. Methods Forty rats were randomly divided into normal control group, model group, LBP group and cisplatin group, with 10 rats in each group. In addition to the normal control group, the other groups were prepared rat brain glioma animal model. LBP group once / d with LBP, Cisplatin group with cisplatin 1 / d, normal control group and model group with saline once / d, the treatment time was 16 weeks. The levels of TNF-α and IL-10 in serum were determined by ELISA. The levels of CD4 ~ + in peripheral blood of each group were detected by flow cytometry CD25 ~ + regulatory T cells (Tr) and CD8 ~ + T cell number. Results Compared with the normal control group, the number of CD4 ~ + CD25 ~ + Tr in the peripheral blood of the model group increased and the number of CD8 ~ + T cells decreased. The serum level of IL-10 increased and the level of TNF-α decreased (P <0.05) ; Compared with the model group, the number of CD4 ~ + CD25 ~ + Tr and the number of CD8 ~ + T cells in LBP group and cisplatin group increased, the serum IL-10 level decreased and TNF-α level increased ). There was no significant difference between the 8th and the 16th week (P> 0.05). Conclusion The immune function of glioma rats decreased, while LBP can enhance the immune function of experimental glioma rats, so as to achieve anti-tumor effect.
