为探讨丝裂霉素(MMC)膀胱癌腔内化疗的作用机制及改善现行临床膀胱癌腔内化疗方案的可能性,以体外模拟膀胱癌腔内化疗的条件,观察低于和类似于临床常用剂量MMC短时处理对膀胱癌EJ细胞的影响,应用光镜、电镜进行形态学观察,并采用末端脱氧核苷酸转移酶介导生物素标记法(TUNEL法)检测发生凋亡的细胞。结果表明,以50%的临床MMC剂量和用药时间能够有效地诱导EJ细胞出现凋亡,达到消灭肿瘤细胞的目的。本实验结果可为拟定更合理的膀胱癌腔内化疗方案提供参考依据。
In order to explore the mechanism of intracavitary chemotherapy of mitomycin C (MMC) bladder cancer and to improve the possibility of intracavitary chemotherapy in the current clinical bladder cancer, the conditions of intracavitary chemotherapy in bladder cancer were simulated in vitro, The effect of short-term dose MMC on bladder cancer EJ cells was observed by light microscopy and electron microscopy. Cell apoptosis was detected by terminal deoxynucleotidyl transferase-mediated biotin labeling (TUNEL). The results showed that EJ cells could effectively induce the apoptosis of EJ cells with 50% clinical MMC dosage and administration time, and achieve the goal of eliminating tumor cells. The results of this experiment can provide a reference for the formulation of a more rational chemotherapy regimen of bladder cancer.
