【目的】探讨系统性红斑狼疮(SLE)患者骨髓基质细胞上清液对造血干(祖)细胞的作用及其机制。【方法】半固体集落培养法观察28例活动期SLE骨髓基质细胞生长及其上清液对正常细胞、自身骨髓干/祖细胞的抑制作用;检测SLE其骨髓基质细胞上清液中TNF-α、IFN-γ的水平;RT-PCR法检测骨髓基质细胞IFN-γmR-NA的表达。【结果】①29%活动期SLE患者骨髓基质细胞的集落数较正常人明显减少(P<0.05)。②体积分数为15%的活动期SLE骨髓基质上清液对正常人骨髓、自身SLE造血干/祖细胞混合集落(CFU-Mix)的形成较对照组具有明显抑制作用(P<0.05)。③活动期SLE患者骨髓基质细胞的上清液TNF-α、IFN-γ的水平高于稳定期SLE组、正常对照组(P<0.05)。④70%活动期SLE患者骨髓基质细胞有IFN-γmRNA的表达,而正常对照则未见有IFN-γmRNA的表达。【结论】活动期SLE患者骨髓基质细胞上清液对正常、自身骨髓造血干(祖)细胞的有抑制作用,这与其基质细胞存在高表达TNF-α、IFN-γ有关。
【Objective】 To investigate the effect and mechanism of bone marrow stromal cell supernatant on hematopoietic stem (progenitor) cells in patients with systemic lupus erythematosus (SLE). 【Methods】 The growth of SLE bone marrow stromal cells in 28 cases and the effect of its supernatant on the proliferation of normal cells and bone marrow stem / progenitor cells were observed by semi-solid colony culture method. The levels of TNF-α , IFN-γ, and the expression of IFN-γmR-NA in bone marrow stromal cells was detected by RT-PCR. 【Results】 ① The percentages of bone marrow stromal cells in 29% active SLE patients were significantly lower than those in normal controls (P <0.05). ② The volume of 15% SLE bone marrow stromal supernatant significantly inhibited the formation of CFU-Mix in normal human bone marrow and SLE hematopoietic stem / progenitor cells (P <0.05). ③ The levels of TNF-α and IFN-γ in supernatant of SLE patients during active phase were higher than those in stable SLE group and normal control group (P <0.05). ④ The expression of IFN-γmRNA in bone marrow stromal cells of 70% of active SLE patients, but no expression of IFN-γmRNA in normal controls. 【Conclusions】 BMSCs supernatant in active SLE patients can inhibit normal and autologous bone marrow hematopoietic progenitor cells, which is related to the high expression of TNF-α and IFN-γ in stromal cells.
