目的探讨中国北方汉族男子脑型肌酸激酶(CKB)基因多态性与耐力训练对心功能影响的关联性。方法102名无训练史的健康男子进行18周有氧耐力训练,训练前后测定左心室结构与功能相关指标;用PCR-RFLP法分析CKB基因启动子上rs3759582和rs3759584两单核苷酸多态性。结果二个多态位点均符合H-W平衡,不存在强的连锁关系。训练前左心结构和功能各指标在CKB两基因位点各组均无显著性差异(P>0.05);训练后rs3759582位点AC组△100WEDD、△150WEDD、△100WSV、△150WSV、△150WSV和△reT增加率显著大于AA组(P<0.05)。△CO和△COI在AC组下降,AA组增加;△150WCO和△150WCOI在AC组上升,AA组下降,且在基因型间均有显著性差异(P<0.05)。训练后各指标变化在rs3759584位点各基因型间均无显著性差异(P>0.05)。结论 CKB基因的rs3759584多态性不是个体心功能耐力训练敏感性的基因标记;rs3759582多态性可能与个体心功能对耐力训练敏感性相关,AC型可能更有利于个体心功能对耐力训练的改善。
Objective To investigate the association of brain-type creatine kinase (CKB) gene polymorphisms and endurance training on cardiac function in Han Chinese men in northern China. Methods Totally 102 healthy men without training history were trained in aerobic endurance training for 18 weeks. Left ventricular structure and function were measured before and after training. Two single nucleotide polymorphisms of rs3759582 and rs3759584 on promoter of CKB gene were analyzed by PCR-RFLP . Results The two polymorphic sites were in accordance with the H-W balance, there is no strong chain relationship. Before training, the indexes of left heart structure and function were not significantly different in each CKB locus (P> 0.05); after training, rs3759582 locus △ 100WEDD, △ 150WEDD, △ 100WSV, △ 150WSV, △ 150WSV and △ reT increased significantly more than AA group (P <0.05). △ CO and △ COI decreased in AC group and increased in AA group. △ 150WCO and △ 150WCOI increased in AC group and decreased in AA group (P <0.05). After training, there was no significant difference in each index between rs3759584 loci (P> 0.05). Conclusion rs3759584 polymorphism of CKB gene is not a genetic marker of sensitivity to individual endurance training. Rs3759582 polymorphism may be related to individual sensitivity to endurance training, and AC may be more beneficial to improve individual endurance training .
