目的观察阳离子多肽MP-1对脓毒症小鼠腹腔巨噬细胞(PMφ)呼吸爆发的影响。方法观察MP-1(3mg/kg)对内毒素(LPS)(20mg/kg)攻击的脓毒症模型小鼠的保护作用;体外培养小鼠PMφ,将MP-1与PMφ在37℃下孵育2h后,用LPS400ng/ml刺激PMφ,采用紫外分光光度法检测并换算得到超氧阴离子(O-2)的量和还原型辅酶Ⅱ(NADPH)氧化酶的活性,荧光法检测过氧化氢(H2O2)浓度。结果MP-1可显著抑制LPS刺激小鼠PMφ产生O-2、H2O2及细胞膜上NADPH氧化酶的活性。结论MP-1对LPS攻击的小鼠有显著的保护作用,其机制可能与MP-1对脓毒症小鼠巨噬细胞呼吸爆发的抑制作用有关。
Objective To observe the effect of cationic polypeptide MP-1 on the respiratory burst of peritoneal macrophages (PMφ) in septic mice. Methods The protective effect of MP-1 (3mg / kg) on sepsis model mice challenged with endotoxin (LPS) (20mg / kg) was observed. PMΦ was cultured in vitro and MP-1 and PMφ were incubated at 37 ℃ After 2h, the PMΦ was stimulated with LPS400ng / ml, and the amount of superoxide anion (O-2) and the activity of reduced coenzyme Ⅱ (NADPH) oxidase were detected by ultraviolet spectrophotometry and detected by hydrogen peroxide )concentration. Results MP-1 could significantly inhibit the production of O-2, H2O2 and the activity of NADPH oxidase on the membrane of mice by LPS stimulation. Conclusions MP-1 has a significant protective effect on LPS-challenged mice, and its mechanism may be related to the inhibitory effect of MP-1 on respiration burst of murine macrophage.
