目的:研究牛膝醇提物含药血清对骨髓间充质干细胞(BMSCs)体外增殖的影响。方法:采用高、中、低剂量牛膝醇提物以及等体积生理盐水对SD级新西兰大白兔进行灌胃,从中提取含药血清,同时采用贴壁筛选法培养兔骨髓间充质干细胞,并以4种不同浓度的含药血清及胎牛血清分别干预第三代骨髓间充质干细胞。于干预后第3、6、9天分别3次采用CCK-8法测定细胞光密度值(OD),比较各组间的差异。结果:高剂量组牛膝醇提物含药血清刺激骨髓间充质干细胞增殖,并促进细胞分化,其OD均值与其他组相比较最高,差异均具有统计学意义(P<0.05)。结论:牛膝醇提物含药血清具有促进BMSCs增殖作用,其中高剂量作用最强。
Objective: To study the effect of Achyranthes bidentata extract-containing serum on proliferation of bone marrow mesenchymal stem cells (BMSCs) in vitro. Methods: SD high grade New Zealand white rabbits were gavaged with high, medium and low doses of Achyranthes bidentata alcohol extract and an equal volume of normal saline. The serum was extracted from the SD rabbits and the BMSCs were cultured by adherence screening The third generation of bone marrow mesenchymal stem cells were treated with four different concentrations of serum and fetal bovine serum respectively. Cell optical density (OD) was measured by CCK-8 method on the 3rd, 6th and 9th day after the intervention respectively, and the differences between the groups were compared. Results: The high dose group of Achyranthes root alcohol extract containing serum stimulated the proliferation of bone marrow mesenchymal stem cells, and promote cell differentiation, the average OD value compared with other groups, the difference was statistically significant (P <0.05). CONCLUSION: Achyranthes bidentata ethanol-containing serum can promote the proliferation of BMSCs, of which the highest dose is the strongest.
