目的探讨自噬标志物——微管相关蛋白轻链3B(MAPLC3B)在人肺鳞癌组织中的表达以及与窖蛋白-1(Cav-1)的关系及它们的临床意义。方法利用量子点免疫荧光组织化学技术检测LC3B和Cav-1蛋白在肺鳞癌组织芯片包括100例肺鳞癌组织和20例非癌性肺组织上的表达;同时在20例肺鳞癌和20例非癌性肺组织中利用定量实时PCR法检测LC3B和Cav-1 mRNA的变化。结果与非癌变肺组织相比,LC3B和Cav-1蛋白和mRNA在肺鳞癌组织的表达均显著降低(P<0.01)。LC3B蛋白阳性表达与男性肺鳞癌患者显著相关,与淋巴结转移相关,但差异无显著性(P>0.05)。肿瘤细胞Cav-1蛋白阳性表达在肺鳞癌TNM分期(P=0.009)和淋巴结有无转移(P=0.027)之间的差异均有显著性,而与其他临床病理参数均无关(P>0.05)。基质Cav-1阳性率仅为10.12%(8/79),与肺鳞癌临床病理参数均无关(P>0.05)。LC3B和Cav-1蛋白在肺鳞癌肿瘤细胞的表达呈显著正相关(P=0.001,r=0.473)。结论自噬基因LC3B和Cav-1的缺失表达可能与肺鳞癌的发生有关,但二者共同阳性表达又可协同促进肿瘤的恶性进展。
Objective To investigate the expression of autophagy marker microtubule-associated protein light chain 3B (MAPLC3B) in human lung squamous cell carcinoma and its relationship with caveolin-1 (Cav-1) and their clinical significance. Methods The expression of LC3B and Cav-1 protein in lung squamous cell carcinoma tissue samples including 100 cases of lung squamous cell carcinoma and 20 cases of non-cancerous lung tissues were detected by quantum dot immunofluorescence histochemistry. In non-cancerous lung tissues, quantitative real-time PCR was used to detect the changes of LC3B and Cav-1 mRNA. Results Compared with non-cancerous lung tissues, the expression of LC3B and Cav-1 protein and mRNA in lung squamous cell carcinoma tissues were significantly decreased (P <0.01). The positive expression of LC3B protein was significantly associated with lung squamous cell carcinoma in patients with lymph node metastasis, but the difference was not significant (P> 0.05). The positive expression of Cav-1 protein in tumor cells was significantly different between TNM staging (P = 0.009) and lymph node metastasis (P = 0.027), but not with other clinicopathological parameters (P> 0.05 ). The positive rate of Cav-1 in stroma was only 10.12% (8/79), which had no correlation with the clinicopathological parameters of squamous cell carcinoma (P> 0.05). There was a significant positive correlation between LC3B and Cav-1 protein expression in lung squamous cell carcinoma (P = 0.001, r = 0.473). Conclusions The loss of the expression of autophagy genes LC3B and Cav-1 may be related to the occurrence of squamous cell carcinoma of the lung. However, the positive expression of both autophagy gene and Cav-1 may synergistically promote the malignant progression of the tumor.
