目的:研究强心通脉颗粒对慢性心衰大鼠心肌组织AngⅡ与血栓烷素B2(TXB2)的影响。方法:以结扎大鼠冠状动脉致其急性心肌梗死后,再进行饥饿及力竭式游泳造成大鼠慢性心衰模型。并在用药6周后,用彩色超声多普勒心动图测量大鼠的心功能,以免疫组化方法测定心肌组织中AngⅡ的含量并用ELESA法测定各组TXB2的数值。结果:大鼠模型组与正常对照组比较,心功能明显降低(P<0.01);AngⅡ、TXB2含量远远高于正常组(P<0.01);中药组和西药组AngⅡ、TXB2含量均低于模型组(P<0.01)。结论:强心通脉颗粒能抑制慢性心衰大鼠RAS系统的激活,降低血清中TXB2的水平,从而能改善慢性心衰大鼠的心功能,为慢性心衰防治及实验研究提供客观依据。
Objective: To study the effect of Qiangxin Tongmai Granule on myocardial AngII and thromboxane B2 (TXB2) in rats with chronic heart failure. METHODS: After ligating the coronary artery in rats to induce acute myocardial infarction, chronic heart failure was induced by starvation and exhausted swimming. After 6 weeks of treatment, the heart function of rats was measured by color Doppler echocardiography. The content of Ang II in myocardial tissue was measured by immunohistochemistry and the value of TXB2 in each group was determined by ELESA method. RESULTS: Compared with the normal control group, the heart function of the rat model group was significantly lower (P<0.01); the content of AngII and TXB2 was much higher than that of the normal group (P<0.01); the contents of AngII and TXB2 were lower in the Chinese medicine group and the western medicine group. Model group (P<0.01). Conclusion: Qiangxin Tongmai Granules can inhibit the activation of RAS system in rats with chronic heart failure and reduce the level of TXB2 in serum, which can improve the heart function of chronic heart failure rats, and provide an objective basis for the prevention and experimental study of chronic heart failure.
