目的观察槲皮素联合伊马替尼对人胃癌SNU-1细胞生长和细胞凋亡的影响。方法以12.5~200.0μmol/L槲皮素和0.125~2.000μmol/L伊马替尼单药以及二者联合用药处理细胞24 h,用噻唑蓝(MTT)法检测细胞增殖,Hoechst 33342染色观察细胞核变化,流式细胞仪检测细胞凋亡情况;用蛋白印迹法检测相关蛋白的表达情况。结果槲皮素和伊马替尼单药均能使细胞活力降低,0.250μmol/L伊马替尼和2 5.0μmol/L槲皮素联合用药对细胞活力具有显著抑制作用,差异有高度统计意义(P<0.01),且可诱导人胃癌SNU-1细胞凋亡;二者联合处理还可以显著诱导半胱氨酸蛋白酶-9(Caspase-9)和半胱氨酸蛋白酶-3(Caspase-3)的上调,差异有高度统计意义(P<0.01)。结论槲皮素联合伊马替尼能抑制胃癌SNU-1细胞的生长并诱导细胞凋亡,其作用机制主要是通过上调Caspase-9和Caspase-3的表达从而启动细胞凋亡来实现的。
Objective To observe the effect of quercetin and imatinib on the growth and apoptosis of human gastric cancer cell line SNU-1. Methods The cells were treated with 12.5 ~ 200.0μmol / L quercetin and 0.125 ~ 2.000μmol / L imatinib monotherapy as well as the combination of both for 24 hours. Cell proliferation was detected by MTT assay. Hoechst 33342 staining was used to observe the cell nucleus The changes of apoptosis were detected by flow cytometry. The expression of related protein was detected by Western blotting. Results Both quercetin and imatinib could decrease cell viability. Combination of 0.250μmol / L imatinib and 2 5.0μmol / L quercetin significantly inhibited cell viability, the difference was statistically significant (P <0.01), and could induce the apoptosis of human gastric cancer SNU-1 cells. The combined treatment could significantly induce the expression of Caspase-9 and Caspase-3 ), The difference was highly statistically significant (P <0.01). Conclusion Quercetin combined with imatinib can inhibit the growth of gastric cancer cell line SNU-1 and induce cell apoptosis. The mechanism is mainly through the up-regulation of the expression of Caspase-9 and Caspase-3 to initiate apoptosis.
