目的:研究藏药二十五味珊瑚丸大鼠长期毒性实验各组织中铜、汞、铅三种重金属元素体内的分布情况。方法:二十五味珊瑚丸0.333,0.667,1.333g(原生药)/kg连续灌胃大鼠三个月,末次给药后分别取高剂量组和对照组6只大鼠的心、肝、脾、肺、肾、脑、胃-十二指肠、结肠(降结肠)及全血进行含量测定。微波消解仪消解组织样品,原子吸收仪测铜、汞、铅含量。结果:高剂量组大鼠各组织的汞含量顺序为:肾>结肠(降结肠)>脾>胃-十二指肠>肺>肝>血>心>脑;其中肝脏、肾脏的汞含量高于对照组;脾、胃-十二指肠、结肠(降结肠)及全血等组织的汞含量均有高于对照组趋势;心、肺、脑等组织的汞含量与对照组比较无显著差异。大鼠高剂量组体内各组织的铜含量顺序为:肾>肝>心>脑>胃-十二指肠>肺>脾>血>结肠(降结肠),各组织的铜含量与对照组比较统计差异不显著。大鼠高剂量组铅含量顺序为:脑>脾>结肠>胃-十二指肠>血>肝>心>肺>肾,其中心脏的铅含量高于对照组(P<0.05);肝、脾、肺、脑、胃-十二指肠及全血等组织的铅含量均有高于对照组趋势;肾、结肠(降结肠)与对照组比较无显著差异。结论:二十五味珊瑚丸灌胃大鼠3个月,汞在肾脏和肝脏可能有一定蓄积性,铅在心脏可能有一定蓄积性,但其生物学意义尚需结合大鼠长期毒性试验给药90天和停药恢复15天的血液细胞学、血清生化、尸体解剖及组织病理学学结果做进一步讨论和分析。
Objective: To study the distribution of three heavy metal elements Cu, Hg and Pb in long-term toxicity experiment of Tibetan medicine Twenty-five Corals. The contents of copper in the tissues of high-dose group were as follows: kidney> liver> heart> brain> stomach-duodenum> lung> spleen> blood> colon (descending colon) Statistical differences are not significant. The levels of lead in the high-dose group were as follows: brain> spleen> colon> stomach-duodenum> blood> liver> heart> lung> The contents of Pb in spleen, lung, brain, stomach, duodenum and whole blood were higher than those in control group. There was no significant difference between kidney and colon (descending colon) and control group. CONCLUSION: Twenty-five coral pills are administered to rats for 3 months. Mercury may accumulate in the kidneys and liver, and lead may accumulate in the heart. However, its biological significance needs to be combined with long-term toxicity test in rats Hematology, serum biochemistry, autopsy and histopathology results for 90 days of drug withdrawal and 15 days of withdrawal from treatment were further discussed and analyzed.
