目的探讨妇科千金片治疗慢性宫颈炎的作用机制。方法将60只SD雌鼠随机分为6组:正常对照组,模型对照组,妇科千金片小、中、大剂量组(1.5、3、6 g/kg)和妇净舒片组。用苯酚胶浆制备大鼠宫颈炎模型,造模结束后观察造模是否成功。各给药组自造模后第4日开始灌胃给药,灌胃容积为10 ml/kg,连续给药21 d,正常对照组和模型对照组给予等容量净水,治疗后,用RT-q PCR检测大鼠宫颈IL-1β、MKP-1 mRNA的表达水平,用Western blot检测大鼠宫颈MKP-1蛋白表达水平。结果与空白组比较,模型组IL-1βmRNA水平明显升高,差异有统计学意义(P<0.05),与模型组相比,用妇科千金片和妇净舒片治疗后IL-1βmRNA水平显著下降,差异有统计学意义(P<0.05)。而与空白组比较,模型组、妇科千金片低、中、高剂量组、妇净舒片组的MKP-1 mRNA和蛋白水平明显升高,差异有统计学意义(P<0.05)。结论妇科千金片可能是通过调控MKP-1 mRNA和蛋白表达而发挥抗炎作用。
Objective To investigate the mechanism of gynecological Qianjin tablets in the treatment of chronic cervicitis. Methods Sixty female SD rats were randomly divided into 6 groups: normal control group, model control group, gynecological Qianjin small, medium and large dose groups (1.5, 3, 6 g / kg) and Fukangshu Tablet group. Compared with the model group, the level of IL-1βmRNA decreased significantly , The difference was statistically significant (P <0.05). Compared with the blank group, the mRNA and protein levels of MKP-1 in the model group, the gynecological Qianjin low, middle and high dose groups and the Fukangshu tablet group were significantly increased (P <0.05). Conclusion Gynecology daughter gold tablets may play an anti-inflammatory role by regulating the expression of MKP-1 mRNA and protein.
