目的本研究旨在探讨miR-1和PIK3CA在非小细胞肺癌(NSCLC)组织中的表达及其临床意义。方法利用RT-qPCR法检测55例NSCLC组织及其癌旁正常组织中miR-1和PIK3CA的表达,并用Western-blot法检测NSCLC组织及其正常组织中PI3K蛋白的表达变化。结果 miR-1在NSCLC原发灶中的表达明显低于在癌旁和正常组织中的表达(P=0.0122),PIK3CA在NSCLC原发灶和癌旁组织中的表达都明显高于在正常组织中的表达(P=0.0446,P<0.0001),且PIK3CA在原发灶中的表达明显高于在癌旁组织中的表达(P=0.0029);miR-1和PIK3CA在术后1年内发生/不发生转移癌组织中的表达差异具有统计学意义(P=0.0369,P=0.0061);PIK3CA的表达与NSCLC组织学类型相关(P=0.0200),但miR-1和PIK3CA的表达与患者的年龄、性别、肿瘤的大小及分化程度无明显相关关系(P>0.05),miR-1的表达与PIK3CA的表达成负相关(r=-0.7096,P<0.05)。结论 miR-1的低表达和PIK3CA的高表达可能和NSCLC的发生及侵袭转移有关,且miR-1和PIK3CA在NSCLC转移过程中相互抑制,可能成为NSCLC治疗的新的潜在靶点。
Objective This study aimed to investigate the expression and clinical significance of miR-1 and PIK3CA in non-small cell lung cancer (NSCLC). Methods RT-qPCR was used to detect the expression of miR-1 and PIK3CA in 55 NSCLC tissues and normal tissues. The expression of PI3K protein in NSCLC tissues and normal tissues was detected by Western-blot. Results The expression of miR-1 in primary NSCLC was significantly lower than that in adjacent normal tissues (P = 0.0122). The expression of PIK3CA in primary NSCLC tissues and adjacent normal tissues was significantly higher than that in normal tissues (P = 0.0446, P <0.0001). The expression of PIK3CA in primary tumor was significantly higher than that in paracancerous tissue (P = 0.0029) (P = 0.0369, P = 0.0061). The expression of PIK3CA was correlated with the histological type of NSCLC (P = 0.0200), but the expression of miR-1 and PIK3CA was not associated with the age of patients (P> 0.05). There was a negative correlation between the expression of miR-1 and PIK3CA (r = -0.7096, P <0.05). Conclusions The low expression of miR-1 and the high expression of PIK3CA may be related to the occurrence and invasion of NSCLC. The inhibition of miR-1 and PIK3CA in NSCLC may be a new potential target for the treatment of NSCLC.
