目的:观察干扰素γ(IFN-γ)对哮喘小鼠肺组织中水通道蛋白5(AQP5)表达的影响。方法:24只清洁级BALB/c小鼠随机分为对照组、哮喘组和IFN-γ组,每组8只,后2组制作哮喘模型,IFN-γ组用IFN-γ治疗。采用免疫组织化学法检测肺组织中AQP5蛋白的表达。结果:哮喘组肺组织AQP5蛋白相对表达量为(0.1186±0.0007),较对照组的(0.2060±0.0005)减低(P<0.01),IFN-γ治疗后AQP5蛋白相对表达量为(0.1479±0.0002),较哮喘组增加(P<0.01)。结论:AQP5蛋白表达下调可能参与了哮喘发病时气道黏液分泌的调节,IFN-γ可上调AQP5蛋白的表达,在哮喘气道黏液分泌中起治疗作用。
Objective: To observe the effect of interferon γ (IFN-γ) on the expression of aquaporin 5 (AQP5) in the lung tissue of asthmatic mice. Methods: Twenty-four clean BALB / c mice were randomly divided into control group, asthma group and IFN-γ group, with 8 rats in each group. The latter two groups were given asthma model. IFN-γ group was treated with IFN-γ. Immunohistochemistry was used to detect the expression of AQP5 protein in lung tissue. Results: The relative expression of AQP5 protein in asthmatic group was (0.1186 ± 0.0007), lower than that in control group (0.2060 ± 0.0005) (P <0.01), and the relative expression of AQP5 protein was (0.1479 ± 0.0002) , More than the asthma group (P <0.01). CONCLUSION: The down-regulation of AQP5 protein may be involved in the regulation of airway mucus secretion during the onset of asthma. IFN-γ may up-regulate the expression of AQP5 protein and play a therapeutic role in the airway mucus secretion of asthma.
