利用混合悬浮培养的方法,构建了一个由重组大肠杆菌的谷胱甘肽生物合成酶系与面包酵母的糖酵解途径组成的ATP再生系统,验证了该系统用于谷胱甘肽(GSH)生物合成的可行性.研究了葡萄糖浓度、细胞量对耦合系统合成GSH 能力的影响,讨论了所构建的ATP再生系统中GSH 合成能力较低的可能原因.
A hybrid regeneration system consisting of the recombinant E. coli glutathione biosynthetic enzyme system and baker’s yeast glycolysis pathway was constructed. The system was used to determine glutathione (GSH) Feasibility of biosynthesis. The effects of glucose concentration and cell mass on the ability of GSH to synthesize GSH were investigated. The possible reasons for the low GSH synthesis ability of the constructed ATP regeneration system were discussed.
