HCV的研究主要集中在以下几个方面:①HCV基因及其产物结构与功能的分析;②HCV的复制机制;③HCV保护性抗原的鉴定及疫苗的研制;④抗HCV药物筛选;上述4个方面的研究都需要HCV动物模型和体外细胞模型。筛选出与原始抗原序列完全相同的线性表位(Liner epitope),又可以筛选出与原始抗原序列不同但功能相似的构像表位(conformational epitope),即模拟表位。这些研究及技术为选取中国HCV流行株T细胞模拟表位和构建细胞模型奠定了基础。理想的细胞模型应具备以下基本条件:①稳定性,即细胞在培养条件下能够稳定地复制和表达;②易于重复;③高水平表达基因产物;④检测方法方便敏感。有研究利用人外周血淋巴细胞建立了HCV感染HCV病毒细胞模型。所以采用人丙型肝炎病毒T细胞模拟表位片段融合基因转染外周血淋巴细胞具有充分依据。

Some studies have used human peripheral blood lymphocytes to establish a HCV infected HCV virus cell model. Therefore, human Hepatitis C virus T cell mimic epitope fragment fusion gene transfected peripheral blood lymphocytes with sufficient basis.
