目的:探讨护骨胶囊对体外培养的大鼠成骨细胞增殖、分化的影响。方法:采用多次酶消化法分离出大鼠颅盖骨的成骨细胞,采用MTT法和碱性磷酸酶法分别检测护骨胶囊对成骨细胞增殖和分化的作用;采用SYBR greenⅠ嵌合的RT-PCR方法检测Runx2 mRNA表达水平。结果:护骨胶囊在浓度0.025~2.5μg/mL对成骨细胞有促增殖的作用,护骨胶囊在浓度0.025~25μg/mL对碱性磷酸酶的活性均有提高的作用,护骨胶囊在浓度25μg/mL下,对Runx2 mRNA水平有上调的作用。结论:护骨胶囊对成骨细胞有促增殖、促分化的作用,可能与上调Runx2 mRNA水平有关。
Objective: To investigate the effect of Hu Gu capsule on the proliferation and differentiation of cultured rat osteoblasts. Methods: Osteoblasts of rat calvaria were isolated by multiple enzymatic digestion. MTT assay and alkaline phosphatase assay were used to detect the effects of Prescription Capsule on proliferation and differentiation of osteoblasts respectively. Chimerism of SYBR greenⅠ RT-PCR method to detect Runx2 mRNA expression levels. Results: The protective effect of Hugu capsule on the proliferation of osteoblasts at the concentration of 0.025 ~ 2.5μg / mL was observed. The protective effect of Hugu capsule on alkaline phosphatase activity at the concentration of 0.025 ~ 25μg / mL, At a concentration of 25 μg / mL, up-regulation of Runx2 mRNA levels was observed. Conclusion: Capsule on promoting proliferation and differentiation of osteoblasts may be related to the up-regulation of Runx2 mRNA level.
