目的:探讨加减抵当汤对IR大鼠血浆t-PA、PAI-1水平及脂肪PAI-1mRNA表达的影响。方法:造模大鼠随机分成正常组、模型组、加减抵当汤高、中、低剂量组、文迪雅组,用发色底物法检测t-PA、PAI-1水平;RT-PCR方法检测加减抵当汤对IR大鼠脂肪PAI-1 mRNA表达的影响。结果:加减抵当汤高、中剂量组能明显升高IR大鼠血浆t-PA水平,降低PAI-l水平(P<0.05和P<0.01),下调PAI-1mRNA的表达。结论:加减抵当汤能升高血浆t-PA,降低PAI-1;并能下调脂肪PAI-1mRNA的表达,可能是其改善IR的机理之一。
Objective: To investigate the effect of decoction and decoction on blood plasma t-PA, PAI-1 and expression of adipose PAI-1 mRNA in IR rats. METHODS: Model rats were randomly divided into normal group, model group, high-, middle-, and low-dose groups of Wentangya Decoction and Wendiya group. The levels of t-PA and PAI-1 were detected by chromogenic substrate method; RT-PCR. Methods The effect of decoction and decoction on the expression of PAI-1 mRNA in adipose tissue of IR rats was detected. RESULTS: Addition and subtraction of the decoction high and middle dose groups significantly increased plasma t-PA levels, decreased PAI-1 levels (P<0.05 and P<0.01), and down-regulated PAI-1 mRNA expression in IR rats. Conclusion: Addition and subtraction of decoction can increase plasma t-PA, decrease PAI-1, and down-regulate the expression of PAI-1 mRNA, which may be one of the mechanisms of improving IR.
