目的用TK基因突变试验和彗星试验检测亚硒酸钠(Na2SeO3)的致突变性和抗突变性。方法设阴性对照组、阳性对照组、抗突变对照组,以及致突变试验组和抗突变试验组各5个剂量组,处理L5178Y细胞3 h,按常规方法进行TK基因突变试验和彗星试验。结果 TK基因突变试验中,6.0~24.0μg/ml Na2SeO3致突变试验组的突变频率(MF)显著高于阴性对照组;0.2~1.6μg/ml Na2SeO3抗突变试验组的突变频率低于丝裂霉素C(MMC)阳性对照组。在彗星试验中,3.0~24.0μg/ml Na2SeO3致突变试验组的平均拖尾率和Olive尾距(OTM)显著大于阴性对照组;0.2~1.6μg/ml Na2SeO3抗突变试验组的平均拖尾率和平均OTM值显著低于重铬酸钾(K2Cr2O7)阳性对照组。结论在不同的剂量范围内,亚硒酸钠可表现出一定的致突变作用和抗突变作用。
Objective To test the mutagenicity and anti-mutagenicity of sodium selenite (Na2SeO3) by TK gene mutation test and comet assay. Methods The negative control group, the positive control group, the anti-mutation control group, and the mutagenicity test group and the anti-mutation test group were respectively treated with 5 doses to treat L5178Y cells for 3 h. TK gene mutation test and comet assay were carried out by conventional methods. Results In the TK gene mutation test, the mutation frequency (MF) in the group of 6.0 ~ 24.0μg / ml Na2SeO3 mutagenicity was significantly higher than that in the negative control group. The frequency of mutation in the group of 0.2 ~ 1.6μg / ml Na2SeO3 anti- C (MMC) positive control group. In comet assay, the average tailing rate and Olive tail length (OTM) of 3.0 ~ 24.0μg / ml Na2SeO3 mutagenicity test group were significantly greater than those of the negative control group. The average tailing rate of 0.2 ~ 1.6μg / ml Na2SeO3 anti-mutation test group And the average OTM was significantly lower than potassium dichromate (K2Cr2O7) positive control group. Conclusions Sodium selenite shows certain mutagenicity and anti-mutagenic effects in different dosage ranges.
