目的:建立黄芪中总皂苷的含量测定方法。方法:采用比色法测定。以5%香草醛-冰醋酸溶液和高氯酸为显色剂,测定波长为530nm,并采用液相色谱-质谱联用技术(LC-MS)验证了测定结果的可靠性。结果:总皂苷测定的线性范围为0.10~1.0mg,r=0.9999;平均回收率为99.8%,RSD为4.1%(n=9)。含量测定结果与LC-MS的测定结果相符。结论:本法简便、准确,可用于黄芪药材总皂苷的含量测定。
Objective: To establish the method of determination of total saponins in Radix Astragali. Method: The colorimetric method was used. The detection wavelength was 530 nm with 5% vanillin-glacial acetic acid solution and perchloric acid as the chromogenic reagent, and the reliability of the determination results was confirmed by liquid chromatography-mass spectrometry (LC-MS). Results: The linear range of determination of total saponins was 0.10 ~ 1.0mg, r = 0.9999; the average recovery was 99.8% with RSD of 4.1% (n = 9). Content determination results and LC-MS measurement results. Conclusion: This method is simple, accurate and can be used for the determination of total saponins of Radix Astragali.
