目的建立松龄血脉康胶囊中葛根及马尾松鲜松叶的UPLC指纹图谱分析方法,为其质量评价累积数据。方法采用RP-HPLC法,以Agilent Eclipse Plus C18 RRHD色谱柱(150 mm×2.1 mm,1.8μm);流动相为0.1%甲酸(A)-乙腈(B),梯度洗脱时间60 min;检测波长250 nm;体积流量0.2 mL.min-1;柱温30℃;进样量1μL,进行指纹图谱研究。同时通过液相色谱-电喷雾离子阱质谱分析方法鉴别部分成分。结果建立了松龄血脉康胶囊的指纹图谱,以葛根素为参照峰,确定了28个共有峰,测定了32批样品,样品指纹图谱相似度均大于99%,共鉴别出25个相关成分。结论所建方法操作简便,分离效果好、稳定性高、重复性好,UPLC法使缩短了检测时间,可应用于松龄血脉康胶囊的质量控制及真伪品鉴别。
OBJECTIVE To establish UPLC fingerprinting method for determination of Pueraria lobata and Masson pine leaves in Songling Capsule of Songling Capsule. Methods The mobile phase was 0.1% formic acid (A) -acetonitrile (B) with gradient elution time of 60 min on an Agilent Eclipse Plus C18 RRHD column (RP-HPLC) with a mobile phase of 150 mm × 2.1 mm and a resolution of 1.8 μm. 250 nm; volume flow rate of 0.2 mL.min-1; column temperature of 30 ℃; injection volume of 1μL, fingerprinting studies. At the same time, some components were identified by liquid chromatography-electrospray ion trap mass spectrometry. Results Fingerprints of Songlingxuemaikang Capsules were established. 28 common peaks were identified with puerarin as the reference peak. 32 batches of samples were determined. The similarity of fingerprints of the samples was over 99%. A total of 25 related components were identified. Conclusion The method is simple and convenient to operate, with good separation effect, high stability and good reproducibility. The UPLC method can shorten the test time and can be applied to the quality control and authenticity identification of Songling Capsule.
