目的:探讨微乳薄层色谱法用于同时分离鉴定三黄颗粒中多种主要成份。方法:以聚酰胺薄膜为固定相,微乳液-甲酸(9:1)作为展开剂,一次点样,同时分离和鉴别三黄颗粒中的多种主要成份;考察了微乳液类型、十二烷基硫酸钠(SDS)、正丁醇、甲酸等因素对分离效果的影响。结果:以微乳液(SDS-正丁醇-正庚烷-水=6.7:15.8:2.5:75.0,g/g)-甲酸(9:1)为展开剂,同时检出三黄颗粒中黄芩甙、绿原酸、大黄素、大黄酚、大黄酸、小檗碱,操作简便,分离效果理想,检出灵敏,且斑点圆而集中,Rf值高的组分亦无扩散现象。结论:建立了一种新型的色谱技术,可以简便、准确、高效地分离和鉴定三黄颗粒中绿原酸、黄芩苷、大黄素、大黄酸、大黄酚、小檗碱。
Objective: To investigate the microemulsion thin-layer chromatography for the simultaneous separation and identification of a variety of main components of Sanhuang granules. Methods: Polyamide film was used as stationary phase and microemulsion-formic acid (9: 1) as developing solvent. The main components of Sanhuang granules were separated and identified. The effects of microemulsion type, dodecane Effect of sodium base sulfate (SDS), n-butanol and formic acid on the separation efficiency. Results: The microemulsion (SDS-n-butanol-n-heptane-water = 6.7: 15.8: 2.5: 75.0, g / g) , Chlorogenic acid, emodin, chrysophanol, rhein, berberine, easy to operate, the separation effect is ideal, the detection is sensitive, and the spot is round and concentrated, and the component with high Rf value does not diffuse. Conclusion: A new chromatographic technique was developed for the determination of chlorogenic acid, baicalin, emodin, rhein, chrysophanol and berberine in Sanhuang granules easily, accurately and efficiently.
