目的验证经内皮微粒蛋白组学筛选出的差异蛋白细胞角蛋白8(keratin8,KRT8)和锚蛋白1(Ankyrin 1,ANK1)与冠心病的相关性。方法选择50例经冠脉造影证实的冠心病患者,以20例造影阴性患者为对照组,收集其外周静脉血,利用酶联免疫吸附实验检测KRT8、ANK1浓度,冠心病患者冠脉狭窄程度使用Gensini评分标准进行评分、分组。结果 KRT8、ANK1在冠心病患者中的浓度明显高于非冠心病组(P<0.05);在冠心病亚组中,Gensini评分高分组与低分组之间KRT8、ANK1浓度差异对比分析,其均P<0.05,且ANK1、KRT8浓度差异与Gensini评分呈正相关。结论 KRT8、ANK1与冠心病有关联,且与冠状动脉病变严重程度呈正相关。
Objective To verify the correlation between differentially expressed keratin 8 (KRT8) and ankyrin 1 (ANK1) and coronary heart disease (CHD) by endothelial microproteomics. Methods Fifty patients with coronary heart disease confirmed by coronary angiography were selected. Twenty patients with negative angiography were selected as the control group. Peripheral venous blood was collected. The concentrations of KRT8 and ANK1 were detected by enzyme-linked immunosorbent assay (ELISA), and the degree of coronary artery stenosis Gensini score to score, group. Results The concentrations of KRT8 and ANK1 in patients with coronary heart disease were significantly higher than those in patients without coronary heart disease (P <0.05). In the subgroup of coronary heart disease, the differences in the concentrations of KRT8 and ANK1 between Gensini high and low groups were compared P <0.05, and the difference of ANK1 and KRT8 concentration was positively correlated with Gensini score. Conclusion KRT8 and ANK1 are associated with coronary heart disease and positively correlated with the severity of coronary artery disease.
